چکیده

It has been a century since the initial discovery of the possible link between viruses and tumors. During the past century, extensive studies have been conducted to understand the relationship between viruses and cancers. The early studies were focused on tumor viruses that do not cause cancers in their natural hosts. These studies provided the basis for the subsequent studies on the six known human oncogenic viruses. These human oncogenic viruses, which include hepatitis B virus, hepatitis C virus, human papillomavirus, Epstein-Barr virus, Kaposi’s sarcoma-associated herpesvirus, and human T-cell leukemia virus 1 are associated with nearly 20% of the human cancer cases. In this chapter, we review the historical aspects of oncogenic virus research, human oncogenic viruses, and the molecular mechanisms of cellular transformation by viruses.

Interaction of viral oncogenic proteins with the Wnt signaling pathway

It is estimated that up to 20% of all types of human cancers worldwide are attributed to viruses. The genome of oncogenic viruses carries genes that have protein products that act as oncoproteins in cell proliferation and transformation. The modulation of cell cycle control mechanisms, cellular regulatory and signaling pathways by oncogenic viruses, plays an important role in viral carcinogenes...

A Review on Gene Involved in Cancer Development and Oncogenic Viruses

Oncogenic viruses are the viruses that cause cancers in their natural hosts or experimental animal systems which are thought to be causative agents of about 15-20% of cancers. They have been broadly classified into the DNA oncogenic viruses and RNA oncogenic viruses based on the nature of the nucleic acid contain within their virion. The oncogenic DNA and RNA viruses that have been identified b...

Cell Transformation by RNA Viruses: An Overview

Studies of oncogenic viruses have made seminal contributions to the molecular biology of cancer. Key discoveries include the identification of viral oncogenes and cellular proto-oncogenes, elucidation of signal transduction pathways, and identification of tumor suppressor genes. The origins of cancer virology began almost exactly one hundred years ago with the discovery of avian sarcoma and acu...

Viruses in cancer cell plasticity: the role of hepatitis C virus in hepatocellular carcinoma

Viruses are considered as causative agents of a significant proportion of human cancers. While the very stringent criteria used for their classification probably lead to an underestimation, only six human viruses are currently classified as oncogenic. In this review we give a brief historical account of the discovery of oncogenic viruses and then analyse the mechanisms underlying the infectious...

DNA Oncogenic Virus-Induced Oxidative Stress, Genomic Damage, and Aberrant Epigenetic Alterations

Approximately 20% of human cancers is attributable to DNA oncogenic viruses such as human papillomavirus (HPV), hepatitis B virus (HBV), and Epstein-Barr virus (EBV). Unrepaired DNA damage is the most common and overlapping feature of these DNA oncogenic viruses and a source of genomic instability and tumour development. Sustained DNA damage results from unceasing production of reactive oxygen ...
